• bace1（β-site app cleaving enzyme），即β-分泌酶，是淀粉样前体蛋白（amyloid precursor protein，app）产生aβ的关键酶。bace1属于ⅰ型跨膜蛋白，由501个氨基酸组成，其中46～460位氨基酸为功能活性区域。bace1通过水解app670met-671asp之间的肽键，生成670个氨基酸的可溶性n末端和包含完整aβ区的app跨膜c端。后者经γ分泌酶进一步酶切形成aβ。最近的研究表明，bace1基因敲除小鼠脑内的aβ水平可降低90%，且其生理表型基本正常，显示了bace1抑制剂在ad治疗中的广阔前景［1］。通过酵母或昆虫表达系统表达活性bace1在最近已见报道［2，3］。然而，由于上述系统表达的蛋白量低，且纯化流程复杂，不适合bace1抑制剂的大规模筛选。而bace1的原核表达可克服以上缺陷，为筛选特异性抑制剂及研究bace1结构与功能关系提供大量材料，为最终从根本上治疗ad带来希望。
