Platelet/endothelial cell adhesion molecule-1 (PECAM-1) is an important target GP in DITP.1 PECAM-1/CD31 is a 130-kDa member of the immunoglobulin (Ig) superfamily and is expressed on the surface of circulating platelets, monocytes, neutrophils, and selective T-cell subsets.2 PECAM-1 has distinctive features of an immunoreceptor based on its genomic structure and the presence of intrinsic immunoreceptor tyrosine inhibitory motifs (ITIMs) in its ligand binding polypeptide.3 PECAM-1 has been shown to contain functional ITIMs within its cytoplasmic domain, and co-ligation of PECAM-1 with the T-cell antigen receptor (TCR) results in tyrosine phosphorylation of PECAM-1, and subsequent recruitment of Src homology 2 domain-containing protein tyrosine phosphatase-2 (SHP-2), and then attenuation of TCR-mediated cellular signaling.4 The standard used in this kit is the product of recombinant gene expression (extracellular region), consisting of 563 amino acids (E28-K590) with a molecular weight of 65.5kDa. Due to glycosylation, its molecular weight changes to 91-100kDa. ScienCell's mouse PECAM-1 ELISA kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. Mouse PECAM-1-specific polyclonal antibodies are pre-coated onto 8 x 12 divisible strips. The mouse-specific detection polyclonal antibodies are biotinylated. The test samples and biotinylated detection antibodies are subsequently added to the wells and then washed with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB is used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changes to yellow after adding acidic stop solution. The intensity of yellow is proportional to the amount of mouse PECAM-1 in the sample that is captured on the strips.
