组织因子途径抑制因子(tissue factor pathway in hibitor, TFPI)是组织因子凝血途径的主要抑制因子, 组织因子途径抑制因子在诸如动脉粥样硬化,血管再狭窄,弥漫性血管内凝血DIC等方面,有一定的研究价值。
TFPI-2也称为胎盘蛋白-5(placenta protein 5,PP5)或细胞基质联合丝氨酸蛋白酶抑制物(matrix associated serine protease inhibitor,MSPI)。
TFPI-2是相对分子质量为:25KDa的丝氨酸蛋白酶抑制物, 由脉管系统细胞(内皮细胞、平滑肌细胞、成纤维细胞)合成,并沉积在这些细胞的细胞外基质里。TFPI-2为广谱丝氨酸蛋白酶抑制剂,能有效地抑制基质金属蛋白酶(MMP)、纤溶酶、胰蛋白酶、糜蛋白酶、组织蛋白酶等多种蛋白水解酶的活性,在维持细胞外基质(ECM)结构完整、抑制肿瘤细胞浸润转移方面起着重要作用。TFPI-2分布广泛,在人肝脏、肾脏、心脏、骨骼肌等正常组织中有着高度表达,而在肿瘤组织中也有一定程度的表达。
TFPI2 may play a role in the regulation of plasmin-mediated matrix remodeling. It inhibits trypsin, plasmin, factor VIIa/tissue factor and weakly factor Xa, but it has no effect on thrombin. TFPI2 is specific to umbilical vein endothelial cells, liver, placenta, heart, pancreas, and maternal serum at advanced pregnancy. This inhibitor contains three inhibitory domains. TFPI2 contains 3 BPTI/Kunitz inhibitor domains.
