Aminoacylase-1 / ACY1抗体背景综述
Aminoacylase 1, also known as ACY1, is a cytosolic, homodimeric, zinc-binding enzyme that catalyzes the hydrolysis of acylated L-amino acids to L-amino acids and acyl group, and plays a general role in the cytosolic breakdown of acetylated amino acids generated during protein degradation. As the first member of the zinc-binding enzyme family, ACY1 is expressed in a wide range of tissue with the highest level in the kidney tubular epithelium, and functions in the catabolism and salvage of acylated amino acids. ACY1 is capable of interacting with sphingosine kinase type 1 (SPHK1), which is involved in promoting cell growth and inhibiting apoptosis of tumor cells. ACY1 has been reported to exhibit reduced or absent expression in SCLC cell lines tumors and RCCs. This enzyme is evolutionarily conserved in fish, frog, mouse, and human, and the amino acid sequence of human ACY1 is 93% identical to that of mouse. Defects in ACY1 are the cause of aminoacylase-1 deficiency (ACY1D), an inborn metabolic disorder exhibiting markedly increased urinary excretion of several N-acetylated amino acids.'
