藻红蛋白标记抗体的方法：1. 巯基化藻红蛋白（PE）的制备；（1）将600μl浓度为15.5mg/ml的盐酸巯醇亚胺加到1.2ml浓度为3.6mg/ml的PE中；（2）将以上混合液和1.2ml pH6.8的PB混合，而后装入透析袋置入50mmol/L pH6.8的PB中于4℃透析，过夜；（3）再换用pH7.5 PB透析6h。每个PE分子中可结合8个巯基；2. 巯基PE-IgG的制备；（1）将30μl浓度为1.1mg/ml的异双功能试剂SPDP的乙醇溶液，加入到700μl
巯基化探针引入到二硫化膜介质上，即可发生巯基-交换反应，然后探针通过二硫键连接到膜上。巯基化探针与处理膜表面的连接反应证明上述反应。巯基-交换反应快速并可定量，唯一弊端是巯基化探针使用前可能形成引物二聚体。并且通过阻隔二硫键形成引物二聚体大的试剂都会干扰免疫探针二硫键形成。许多能用来交联的化学试剂可应用于巯基和胺基异源双功能连接。通常情况下，应用到巯基化核苷连接的常用试剂有NHS、MBS及SPDP.前面提到的巯基吡哚连接中二硫键的大多是SPDP催化形成，然而，二硫键易形成链内连接而不是在探针和链
