【背景资料】：20S immunoproteasome: High integrity, high purity human cell line derived 20S immunoproteasome complex. Characterised by SDS-PAGE and fluorogenic peptide substrate activity assay to determine 20S immunoproteasome purity and chymotrypsin-like, trypsin-like and caspase-like activities. Degrades peptide substrates but not polyubiquitinated proteins (ubiquitin independent activity). Background: When cells are stimulated with pro-inflammatory cytokines, such as interferon gamma (IFN-ϒ), most of their constitutively expressed proteasomes are replaced with immunoproteasomes, resulting in alteration of proteasomal cleavage preference, optimisation MHC class I molecule peptide ligand generation, cytokine profile modification and influencing of T-helper cell differentiation. The immunoproteasome has also been reported to play a role in T-cell survival, auto-immune and pro-inflammatory diseases and in virus induced neuroinflammation. During immunoproteasome assembly, the constitutive catalytic subunits (β1, β2, β5) are replaced by the inducible subunits beta1i (β1i, LMP2), beta2i (β2i, MECL-1) and beta5i (β5i, LMP7), resulting in changes in proteasome specificity. β1i shows greater chymotrypsin-like substrate specificity when compared to its standard counterpart, β1, whereas the caspase-like activity is reduced and the trypsin-like activity elevated. Pure standard proteasomes are found, for instance, in erythrocytes, whereas spleen tissue contains almost exclusively immunoproteasomes. Most other tissues and cells contain varying mixtures of both standard and immunoproteasomes, and, additionally, intermediate-types of proteasomes; each of which subpopulations can be further separated into two or more subtypes all exhibiting slightly different specific activities.
