【背景资料】：Key proteasome associated deubiquitinating enzyme (DUB). Recombinant untagged protein. Mammalian proteasomes are associated with three deubiquitinating enzymes (DUBs): RPN11, UCH37 and USP14. UCH37 andUSP14 associate reversibly with the proteasome, whereas RPN11 is a stoichiometric subunit. These enzymes reside on the 19S regulator and remove ubiquitin from substrates before substrate degradation. The activity of RPN11 on the substrate’s ubiquitin chain is thought to be delayed until the proteasome is committed to degrading the substrate. In contrast, USP14 and UCH37 can attack ubiquitin chains independently of commitment to substrate degradation, shortening or trimming chains rather than removing them completely, possibly facilitating proteasomal discrimination between long and short polyubiquitin chains. USP14 has been shown to inhibit the proteasomal degradation of ubiquitin–protein conjugates both in vitro and in cells, a process that can be disrupted by the USP14 specific inhibitor IU1 (VB2240) resulting in proteasome activation. As such USP14 presents a potentially significant therapeutic target for stimulation of protein degradation via proteasome activation. USP14 is itself activated by association with the 26S proteasome and shows minimal deubiquitination activity in the absence of proteasome binding.
