【背景资料】：BAG1 and BAG3 are co-chaperone protein members of the Bcl-2-associated athanogene (BAG) protein family and have been shown to participate in a wide variety of cellular processes including signalling, transcription, stress response, and apoptosis. Under physiological conditions Hsp70 mediated quality control of misfolded proteins is mainly achieved by BAG1-dependent proteasomal degradation of polyubiquitin modified proteins. During ageing or under conditions of acute stress, involving accumulation of misfolded and aggregated proteins, a switch in co-chaperone expression from BAG1 to BAG3 results in upregulation of autophagic degradation through targeting by the ‘autophagy receptor’ protein p62. This suggests that the BAG3 / BAG1 ratio regulates the decision between proteasomal versus autophagic degradation. BAG3 has also been shown to participate in the selective engulfment of ubiquitinated proteins, a process termed chaperone-assisted selective autophagy or CASA, by facilitating the association of the chaperone machinery, specifically BAG3 / Hsp70 / HspB8 complex, with p62 and thus targeting the substrates to the autophagic machinery for lysosomal degradation. Surprisingly, the ubiquitin ligase CHIP, known for its involvement in proteasomal degradation, also participates in CASA and interacts with the BAG3 chaperone complex.
