做了快一年的SYBR G的定量实验了,有一些心得,写出来大家交流下. 也是今天在学校的BBS上有人推荐这里的,觉得很不错. 我们用的机器是ABI 7900,我自己比较喜欢的试剂是ABgene的QPCR sybr ROX MIX. 2*试剂我一般再进行稀释一倍后使用,反应总体系是10ul. 用SYBR G来做的话,引物非常重要. 我设计引物使用Primer 5,一般选择上游引物跨最后和最后第二个外显子,下游引物在最后一个外显子中,实在找不到,可以前移,但是一定保证至少一条
