Cyclopamine

Biological Activity

Cyclopamine is a naturally-occurring steroid alkaloid extracted from the corn lily (Veratrum californicum),which specifically inhibits hedgehog signaling pathway (Hh) by influencing the balance between the active and inactive forms of the Smoothened (Smo) protein ^[1].It has been demonstrated that the inhibitory effect of cyclopamine results from its direct binding to the Smo heptahelical protein bundle ^[2].In vitro assays for a panel of nine pancreatic adenocarcinoma (PAC) cell lines (HPA F-2, Panc 10.05, Panc 8.13, Panc 2.03, AsPC-1, CFPA C-1, Panc-1, BxPC-3, S2013), cyclopamine significantly suppresses cell proliferation with IC₅₀ ranging from 8.79 to 45.09 μM ^[3]. Cyclopamine depletes stem-like cancer cells in glioblastoma and blocks tumor engraftment ^[4]. Cyclopamine acts as an useful tool in studying the role of Hh signaling in the process of tumorigenesis and cancer development, such as medulloblastoma ^[5] and basal cell carcinoma ^[6].
