规格: DA1010-3 DA1010-10 溶液 A: 3ml 10ml 溶液 B: 3ml 10ml 保存: -20℃避光密闭保存,一年有效。避免反复冻融。短期可2-8℃保存。产品简介: 二氨基联ben胺(Diaminobenzidine,DAB)是过氧化物酶(POD)的底物,DAB显色液主要用于免疫过氧化物酶法,适用于辣根过氧化物酶HRP标记的免疫印记或免疫组化反应。过氧化物酶使底物发生反应,于反应部位产生棕色沉淀物。免疫印记可直接在膜上显示条带。免疫组化标本显色时间一般为室温5-20分钟,随后在显微镜下观察显色情况,显色充分后应将标本及时脱水封固。其终产物可直接在光镜下观察,也可经OsO4处理后,增加反应产物的电子密度,用于电镜观察。操作说明:
1. 对于组织切片或蛋白质印记膜,在与辣根过氧化物酶(HRP)标记的抗体或其它形式的探针孵育后,用适当洗涤液洗涤3-5次,每次3-5分钟。
2. 取900ul PBS(或双蒸水),加入50ul溶液A,50ul溶液B混合均匀,即配成DAB工作液。 如需要更大体积工作液,可按比例放大。此溶液必须现用现配,配好后避光保存,1小时内使用,过期后请将剩余的液体废弃。3. 向组织切片或膜上加入适量DAB工作液,确保能充分覆盖样品。
4. 室温避光孵育3-10 分钟或更长时间,避免光照,直至显色至预期深浅。
5. 去除DAB染色工作液,用蒸馏水洗涤2-3次即可终止显色反应。
6. 对于组织切片或细胞样品,显色反应终止后,可对其进行其他染料复染。对于膜,显色反应终止后,可以室温晾干避光保存。注意事项:
1.DAB溶液应低温密封保存。如有结晶析出,应确保结晶完全溶解再行使用;
2.显色工作液应现用现配,新鲜配制的工作液应为无色或浅棕色,如颜色过深,请勿使用;
3.显色时间严格控制,根据情况调整,以免显色过度。固相膜显色数小时后即会褪色,不能永久存在;
4.DAB为可疑致癌物,请采取必要的防范措施。操作时应戴手套,尽量避免与皮肤接触,用后及时彻底冲洗,接触DAB的实验用品最好经洗液浸泡24h后使用。

液界 liquid junction 指二种浓度或不同组成的电解质的接触面。液界可简单地把比重轻的溶液由重叠在重液之上而形成,但随着时间的进行,由于扩散或对流等可多少出现液体的混合。为防止混合可采取放置多孔质的膜以延缓扩散,或不断流动液体更新液界等方法。在液界上可产生液界电位( liquid junction potential, EL )。比较简单的是在浓度不同的同一种一价的电解质溶液间或浓度相同并且具有共同的离子的不同的电解质溶液间的情况下,液界电位的大小与液界的混合
