A simple, reliable and accurate method for the measurement of L-rhamnose in plant extracts, culture media/supernatants and other materials.

INTRODUCTION:

L-Rhamnose occurs naturally in the L-form and is commonly
present as a component of the carbohydrate moiety of eukaryotic
glycoproteins and in plant cell wall polysaccharides. The most
abundant occurrence of L-rhamnose is within the pectic fraction
of plant cell wall polysaccharides particularly as a component of
rhamnogalacturonan I, which consists of the repeating disaccharide
unit [1,4)-α-D-GalpA-(1,2)-α-L-Rhap-(1,4]-, and to a lesser extent in
rhamnogalacturonan II.
L-Rhamnose is commonly used as a non-metabolisable marker
along with lactulose for dual-permeability testing in the diagnosis of
intestinal diseases such as Crohn’s disease or coeliac disease.
This kit (K-RHAM) is suitable for the specific measurement of
L-rhamnose.

KITS:

Kits suitable for performing 50 assays in manual format (or 580
assays in auto-analyser format or 500 assays in microplate format) are
available from Megazyme.The kits contain the full assay method plus:
Bottle 1: Buffer (28 mL, pH 9) plus sodium azide
(0.02% w/v) as a preservative.
Stable for > 2 years at 4°C.
Bottle 2: NAD+ plus ATP. Freeze dried powder.
Stable for > 5 years at -20°C.
Bottle 3: Rhamnulokinase, rhamnulose 1-phosphate aldolase
plus lactaldehyde dehydrogenase suspension
(4.5 mL).
Stable for > 2 years at 4°C.
Bottle 4: Rhamnose isomerase suspension (1.5 mL).
Stable for > 2 years at 4°C.
Bottle 5: L-Rhamnose (5 mL; 0.5 mg/mL).
Stable for > 2 years at 4°C.
