| 描述 | Hygromycin B (Hygrovetine) is an aminoglycoside antibiotic that inhibits protein synthesis in both prokaryotic and eukaryotic cells by interfering with translocation and causing mistranslation by the 70S ribosome. It is commonly used for the selection of cells transfected with the hph or hyg resistance gene. |
| 体外活性 | 方法：小鼠 PDAC 细胞 14387T 用 lentiCRISPRv2 hygro 慢病毒转染细胞，48 h 后，将转染的细胞在含有 Hygromycin B (500μg/mL) 的新培养基中培养两周，筛选转染成功的细胞。 结果：Hygromycin B 筛选出了慢病毒成功转染的细胞。[1] 方法：大肠杆菌用含 Hygromycin B (0-8 μg/mL) 的培养基培养，使用 Klett-Summerson colorimeter 监测细胞生长。 结果：Hygromycin B 以浓度依赖的方式减少了活细胞的数量并增加了倍增时间。Hygromycin B 在 20 μg/mL 浓度下抑制 50% 的活细胞数，在 25 μg/mL 浓度时生长速率减半。[2] |

摘要：用80％乙醇溶液浸提饲料中的越霉素A，浓缩后分别用薄层色谱法对6种展开剂、3种薄层板类型和3种显色剂进行了筛选。研究结果表明，用8O％乙醇溶液提取越霉素A、以氯仿：甲醇：氨水：水=1：4：2：2为展开剂、在碱性硅胶G薄层板上展开、用0.25%茚三酮乙醇溶液显色，能将越霉素A与其他活性成分在薄层板上分离，越霉素A的Rf值为0.47。该法的最低检测浓度为0.04 mg／ml，最低检出限为0.2μg。该方法简单，易操作。 关键词：饲料；越霉素A；薄层色谱法 ．
