| 描述 | Floxuridine (FUDR) is an antimetabolite, floxuridine inhibits thymidylate synthase, resulting in disruption of DNA synthesis and cytotoxicity. |
| 体外活性 | Floxuridine对PEPT1的亲和力高于相应的5'-O-单氨基酸酯前药。[1] Floxuridine与Leucovorin联用能显著增强对人类T淋巴母细胞白血病细胞生长的抑制效果，具有协同抑制作用。[2] Floxuridine显著抑制[(3)H]-肌苷和[(3)H]-腺苷的摄取（控制组的60-70%）。[3] Floxuridine通过抑制细胞增殖，在36天后相对于未处理的对照细胞减少了超过50%，但细胞数量相较于初始细胞密度仍增长了四倍。[4] Floxuridine在体外对人类Tenon囊成纤维细胞的增殖具有延长效应。[5] Floxuridine（FUDR）由于其短的半衰期、陡峭的剂量反应曲线、高的全身清除率和高的肝脏提取率，是肝动脉灌注（HAI）的理想化合物。 |
