| 描述 | Givinostat hydrochloride (ITF2357 hydrochloride) is an inhibitor of HDAC with IC50 of 198 and 157 nM for HDAC1 and HDAC3, respectively. Givinostat hydrochloride exhibits anti-inflammatory, anti-angiogenic, and antineoplastic activities. |
| 体外活性 | Givinostat hydrochloride以浓度依赖的方式抑制JS-1细胞增殖，当浓度达到或超过500 nM时，显著抑制JS-1细胞增殖。与未经LPS处理的组相比，联合使用Givinostat hydrochloride≥250 nM与LPS处理的组细胞抑制率有显著差异[3]。Givinostat hydrochloride在25, 50,及100 nM浓度下可减少IL-1β分泌超过70%。此外，Givinostat hydrochloride能抑制TLR激动剂刺激的PBMCs中IL-6的产生，以及IL-12与IL-18联合作用下的IL-6产生。在50 nM时，IL-6分泌减少至50%，而在100和200 nM时，IL-6分泌没有减少[4]。 |
