| 描述 | Amiodarone hydrochloride is an anti-anginal agent and a class III antiarrhythmic drug that inhibits potassium channels (including wild-type hERG channels and their tail currents, with an IC₅₀ of approximately 45 nM) as well as voltage-gated sodium channels. This leads to prolonged action potential duration in ventricular and atrial myocytes, resulting in reduced heart rate and vascular resistance. It activates ERK1/2 and p38 MAPK signaling pathways in fibroblasts, promoting cell proliferation and myofibroblast differentiation. Amiodarone hydrochloride is widely used in the study of supraventricular and ventricular arrhythmias and can also be used to establish pulmonary fibrosis animal models. |
| 体外活性 | Amiodarone（AM）通过5'-脱碘作用（5'DI）抑制从甲状腺素（T4）到三碘甲状腺原氨酸（T3）的细胞内转化,而不影响从T4到逆转T3（rT3）的细胞内转化.1.25-25 mg/kg Amiodarone在房室结和麻醉犬中,导致窦率的减少,房室结的有效和功能不应期的延长,以及频率依赖的传导延迟.每天50 mg/kg,i.p. 3-4周 Amiodarone在兔的心室肌细胞中,导致iK和ito的电流密度显著降低,而不会影响ICA和IK1密度. |
