| 描述 | (-)-Huperzine A (HupA) , an active Lycopodium alkaloid extracted from the traditional Chinese herb, is a potent, selective and reversible acetylcholinesterase (AChE) inhibitor and has been widely used in China for the treatment of Alzheimer's disease (AD). |
| 体外活性 | 在动物模型和AD患者中,(-)-Huperzine A可改善学习和记忆障碍.大鼠处死30 min后,与对照组使用生理盐水相比,(-)-Huperzine A明显抑制皮质,纹状体,海马区,内延髓,侧隔核,小脑和丘脑中的AchE活性. |
| 体内活性 | 与他克林,毒扁豆碱,加兰他敏和卡巴拉汀相比,(-)-Huperzine A在对AChE活性的抑制更强,但对BuChE的效果最差。(-)-Huperzine A使细胞免受谷氨酸盐、过氧化氢、β-淀粉样蛋白缺血和星形孢菌素诱导的细胞毒性及凋亡。(-)-Huperzine A通过调节凋亡蛋白Bcl-2、Bax、p53和caspase-3的表达,减轻氧化应激,保护线粒体,使神经生长因子及其受体上调及干扰淀粉样前体蛋白代谢等一系列过程实现上述保护作用。(-)-Huperzine A会优先抑制四聚体AChE（G4形式）。 |
