Thymidine, a naturally occurring deoxyribonucleoside and the direct precursor of dTTP, is classically exploited as a reversible DNA-synthesis inhibitor: when supplied exogenously at high concentration it provokes an intracellular imbalance of deoxynucleotide pools that stalls replication forks, activates the ATR–CHK1 checkpoint, suppresses Cyclin E/A-CDK2 activity and thereby synchronizes cultured mammalian cells at the G1/S boundary; upon removal the block is rapidly relieved, allowing synchronous entry into S-phase with minimal cytotoxicity, making thymidine double-block the most widely used protocol for cell-cycle synchronization in molecular and cancer biology.
