| 检测原理 | The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Human KRT15. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Human KRT15. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Human KRT15, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Human KRT15 in the samples is then determined by comparing the OD of the samples to the standard curve. |

在目前的以HRP作为标记酶的商品ELISA试剂盒中，如以TMB为底物，则提供的底物为A和B两瓶应用液；如以OPD为底物，则试剂盒提供OPD片剂或粉剂，临用前配制。一般商品试剂盒显色反应条件为37℃或室温反应15~30min。从理论上说，37℃30min才可以使HRP的底物催化反应完全，尽管在最初的10min内，绝大部分催化反应即可完成。因此，为使弱阳性样本孔能有充分的显色，建议在37℃下反应25~30min后，终止反应比色测定。此外，在加入底物开始显色反应前，最好是先检查一下底物溶液的有效
