| 检测原理 | The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Human IDUa. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Human IDUa. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Human IDUa, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Human IDUa in the samples is then determined by comparing the OD of the samples to the standard curve. |

在临床检验中一般采用商品试剂盒进行测定。前文（2.2）已述，ELISA中有三个必要的试剂：免疫吸附剂、结合物和酶的底物等。完整的ELISA试剂盒包含以下各组分：（1）已包被抗原或抗体的固相载体（免疫吸附剂）；（2）酶标记的抗原或抗体（结合物）；（3）酶的底物；（4）阴性对照品和阳性对照品（定性测定中），参考标准品和控制血清（定量测定中）；（5）酶联物（结合物）及标本的稀释液；（6）洗涤液；（7）酶反应终止液。一、免疫吸附剂已包被抗原或抗体的固相载体在低温（2~8℃）干燥的条件下一般可保存6个月
