4. Incubate with primary antibody in PBTN for 1 hr. to overnight. 5. Wash tissue for 30 min. to overnight in PBS. 6. Incubate for 1 hr. to overnight with HRP-conjugated secondary diluted 1:100 in PBTN. 7. Wash 45 min. with 3 changes

in 1 ml 0.1% BSA in tri-PBS, for 5, 10, 15, 20, and 25 minutes, respectively. 10. Replace last wash with 445 ul Tri-PBS, 5 ul 10% BSA, and 40 ul NGS, rock for 30 minutes. 11. Add 10 ul HRP conjugate secondary antibody? and rock for 90 minutes
