. Incubate these membrane sections with secondary antiserum for 1 hour at room temperature (For detecting human antibodies, we use Sigma #A-3150: goat anti-human IgG alkaline phosphatase conjugate, diluted 1:1000) Wash membranes in TBST for 5 minutes
, incubate for 30 min at 37°C. ( 5g powdered milk in 95 ml PBS)5 Rinse with PBS 3x.6 Add 50 µl of antibody, incubate 30 min at 37°C.7 Rinse with PBS 3x.8 Add 50 µl of goat anti-mouse IgG conjugate with alkaline phosphatase.(1 µl of conjugate in 4 ml of PBS

Dilute 1 µL fresh streptavidin-Alexa Flour 555 in 1 mL PBS (1:1,000). Spin the dilute antibody at high speed for 30' to remove particulates. Add supernatant to specimen and incubate 30' at room temp. Remove antibody and add 1 mL PBS. O.K
