25 ug total protein per lane of various lysates probed with CD3D monoclonal antibody, unconjugated (orb1499380) at 1:5000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at r.t. for 60 min.
ICC staining of CD3D in Jurkat cells (green). Formalin fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 10 minutes at room temperature and blocked with 1% Blocker BSA for 15 minutes at room temperature. Cells were probed with the primary antibody (orb1499380, 1/50) for 1 hour at room temperature, washed with PBS. Alexa Fluor®488 Goat anti-Rabbit IgG was used as the secondary antibody at 1/1000 dilution. The nuclear counter stain is DAPI (blue).
Fixative-fixed, paraffin embedded Human Spleen, Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min, Antibody incubation with CD3D Monoclonal Antibody, Unconjugated (orb1499380) at 1:300 overnight at 4°C, followed by conjugation to the SP Kit (Rabbit) and DAB staining.
Fixative-fixed, paraffin embedded Human Tonsil, Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min, Antibody incubation with CD3D Monoclonal Antibody, Unconjugated (orb1499380) at 1:300 overnight at 4°C, followed by conjugation to the SP Kit (Rabbit) and DAB staining.
