检测原理
肌酐在肌酐氨基水解酶作用下水解为肌酸,进一步经肌酸脒基水解酶作用生成肌氨酸和尿素。肌氨酸在肌氨酸氧化酶催化下产生过氧化氢,后者在过氧化物酶作用下与4-氨基安替比林和N-乙基-N-磺丙基-间`甲`苯`胺反应生成醌亚胺,在546 nm处具有特征吸收峰,吸光度值与肌酐浓度成正比。

Application

Assay Principle
Creatinine is hydrolyzed to creatine by creatinine aminohydrolase, which is further acted upon by creatine amidinohydrolase to produce sarcosine and urea. Sarcosine is catalyzed by sarcosine oxidase to generate hydrogen peroxide. The latter, in the presence of peroxidase, reacts with 4-aminoantipyrine and N-ethyl-N-sulfopropyl-m-toluidine to form quinoneimine, which exhibits a characteristic absorption peak at 546 nm. The absorbance value is proportional to the creatinine concentration.
