Assay Principle

Free hydroxyproline (HYP) is released from the sample via hydrolysis. Under the action of Chloramine T, it undergoes oxidation. The oxidation product then reacts with p-dimethylaminobenzaldehyde (DMAB) to develop color, forming a red compound with a characteristic absorption peak at 560 nm. The HYP content in the sample can be accurately calculated by measuring the absorbance at this wavelength.
