CheKine™ Micro Pectinase Activity Assay Kit is a highly sensitive colorimetric tool designed for the quantitative determination of pectinase activity in a variety of sample types, including plant tissues, bacteria, fungi, and cell culture supernatants. This kit measures the absorbance change at 540 nm resulting from the reddish-brown product formed by the reaction between reducing galacturonic acid (generated from pectin hydrolysis by pectinase) and DNS reagent, enabling accurate assessment of enzymatic activity levels.

Principle

Pectinase catalyzes the decomposition of pectin to produce galacturonic acid. The reducing aldehyde group of galacturonic acid reacts with 3,5-dinitrosalicylic acid (DNS) to develop a color, forming a reddish-brown compound with a characteristic absorption peak at 540 nm. By measuring the absorbance change at this wavelength, pectinase activity can be precisely calculated.
