Sheep anti Human fibronectin antibody detects human fibronectin, a multi-domain protein involved in the connection of cells to the extracellular matrix. Fibronectin is mostly found in dimeric form, consisting of two nearly identical polypeptide chains linked by a pair of C-terminal disulfide bonds. Each fibronectin monomer has a molecular weight of 230–250 kDa. Fibronectin can bind cell surfaces and various compounds including collagen, fibrin, heparin, DNA, and actin. Fibronectin is present in the body in two forms; plasma fibronectin (soluble dimeric form) is secreted by hepatocytes, whilst cellular fibronectin (dimeric or cross-linked multimeric forms), made by fibroblasts, epithelial and other cell types, is deposited as fibrils in the extracellular matrix.
Fibronectin is involved in cell adhesion and migration processes including embryogenesis, wound healing and blood coagulation (Schwarzbauer and DiSimone, 2011) as well as pathologies such as metastasis (Van Obberghen-Schilling et al. 2011).
Sheep anti Human fibronectin antibody recognizes fibronectin as a single band of approximately 260 kDa in a range of human cell lysates by western blotting under reducing conditions. Cross reactivity with rodent cell line lysates is also observed.
