PUMA (p53 upregulated modulator of apoptosis), also known as Bcl-2-binding component 3 (BBC3), is a pro-apoptotic protein belonging to the BH3-only subgroup of the Bcl-2 family. Encoded by the BBC3 gene, PUMA is primarily regulated by the tumor suppressor protein p53 and plays a crucial role in mediating apoptosis in response to various cellular stresses, including DNA damage and oncogenic signals. Upon activation, PUMA binds to anti-apoptotic Bcl-2 family members, such as Bcl-2, Bcl-xL, and Mcl-1, neutralizing their function and thereby promoting the activation of pro-apoptotic proteins Bax and Bak. This interaction leads to mitochondrial outer membrane permeabilization, release of cytochrome c, and activation of the caspase cascade, culminating in programmed cell death. PUMA's function is critical in maintaining cellular homeostasis and has significant implications in cancer biology, as its dysregulation can contribute to tumorigenesis and resistance to therapy.​
