Podocin is a kidney-specific integral membrane protein encoded by the NPHS2 gene. It is predominantly expressed in podocytes—specialized epithelial cells lining the glomerular capillaries—and is a critical component of the slit diaphragm, a structure essential for the kidney's filtration barrier. Podocin interacts with proteins such as nephrin and CD2AP, contributing to the assembly and maintenance of the slit diaphragm complex. Mutations in the NPHS2 gene can disrupt podocin function, leading to conditions like steroid-resistant nephrotic syndrome, characterized by proteinuria and progressive kidney disease.
