The assay uses a SARS-CoV-2 Mpro-specific fluorogenic substrate containing a cleavage site found between nsp4 and nsp5. SARS-CoV-2 Mpro cleaves this substrate separating an EDANS fluorophore from a Dabcyl quencher generating free EDANS, which can be easily quantified using a fluorescence plate reader at excitation and emission wavelengths of 340 and 490 nm, respectively.
