Concentrated shRNA lentiviruses were obtained by precipitating viral containing supernatants with PEG6000 (Solarbio).

Lentivirus production and generation of clonal cell lines.（病毒上清液与PEG 6000按2:1的比例混合，病毒在4°C储存12 h后浓缩；PBS重悬病毒）

After 60 h of transfection, virus supernatant was harvested and mixed with PEG 6000 (Solarbio) in a ratio of 2:1.    The virus was then concentrated after 12 h storage at 4°C. Finally, the virus precipitation was resuspended in PBS and stored at -80°C.

Plant（ YM21棉花幼苗，10% PEG6000 ，干旱胁迫）

The 300 mM NaCl (Solarbio, Beijing, China) solution and 10% PEG6000 (Solarbio, Beijing, China) solution were used to simulate salt stress and drought stress, respectively.  The leaves and roots were collected at 0, 3, 6, 12 and 24 h under different abiotic stresses, and the samples were quick-frozen in liquid nitrogen.  To analyze the tissue-specific expression level of GhTIP1;1-like in TM-1, roots, stems and leaves were collected from seedlings, and buds, sepals, petals, stamens and fiber samples.
