文献和实验Cy5.5激发波长为675nm,不能被红色激光器所激发,也就不存在Cy5.5对APC的干扰,所以用PE-Cy5.5对APC计划没有串色。因此,可能需要牺牲个别荧光染料的亮度来避免荧光渗漏以及分辨率下降。 要通过单标对照进行补偿调节: 所以尽量选择光谱重叠度小的荧光染料,如FITC/PE-Cy7; 选择不同激光激发的荧光染料,如FITC/APC, PE/APC。 4) 尽量避免偶联荧光染料使用带来的假阳性 随着颜色的数量不断增加,实验中可能会用到串联荧光染料
