科研界有几大「神药」——阿司匹林、二甲双胍、姜黄素。其中,二甲双胍以其多种神奇功效,成为当之无愧的「顶流」之一。它既能降血糖、治肥胖,也能抗衰老、防癌症、保护心血管,每隔一段时间就会有新作用被研究人员发现。 自 1957 年,二甲双胍在英国最早被批准应用于临床治疗,如今已被用于治疗糖尿病 60 余年,是治疗糖尿病的一线药物。然而,「是药三分毒」,即便是被称为「神药」的二甲双胍,也存在部分副作用,如胃肠道不良反应,影响维生素 B12 吸收等。 如今,糖尿病越来越多地发生在育龄人群当中
IkB-a突变体阻断NF-kB活化:优点,靶点明确;缺点,抑制效率不如RNAi 4、抑制剂(PDTC、SN50等):优点,应用方法简单;缺点,特异性不强,且作用靶点各异(见下面键接),还可能存在细胞特异性。在此需要指出的是,一些传统药物如糖皮质激素、水杨酸类、砷剂和全反式维甲酸等以及姜黄素、大豆异黄酮等天然化合物经证实也是NF-kB抑制剂;蛋白酶抑制剂PSI、PS-341等可阻止IkB-a磷酸化等,从而也可实现抑制NF-kB活化。 各抑制剂的机制可参见下帖: http
