CD40 ligand (CD40LG) is a type II transmembrane glycoprotein belonging to the TNF superfamily. Initially discovered on the surface of CD4+ T cells following T cell receptor activation, CD40LG exists in both membrane-bound and soluble trimeric forms. CD40LG serves as the ligand for CD40, and its binding initiates signal transduction in cells expressing CD40LG. CD40LG is a differentiation antigen expressed on the surface of T cells. In the presence of cytokines, it stimulates B cell proliferation and the secretion of all immunoglobulin isotypes. CD40LG has been shown to induce cytokine production and tumor cell killing. It is also known by the aliases CD154, HIGM1, IGM, IMD3, T-BAM, TNFSF5, TRAP, and gp39.
