| 产品简介(Background) | Beyotime's Recombinant Traptavidin is a mutant of streptavidin, which was expressed in E. coli and purified. Compared to streptavidin, Traptavidin has about 10-fold lower off-rate from biotin (about 10-fold higher binding activity to biotin), increased mechanical and thermal stability [1]. The molecular mass for native Traptavidin is ~52kDa per tetramer, and denatured Traptavidin gives a major band at ~13kDa. |

Traptavidin和Streptavidin相比具有更低的生物素解离速率、更高的热稳定性。在温度为37ºC、pH7.4的反应条件下，利用Biotin-4-Fluorescein测定Traptavidin和Streptavidin与生物素的解离速度，可见Traptavidin与生物素的解离速率(Dissociation %)明显更低，因此，Traptavidin与生物素的结合更为稳固(图1A)。Traptavidin加热至75ºC时仍能保持具有活性的四聚体(Tetramer)，加热至80ºC以上才逐步解聚成失去活性的单体(Monomer)，而Streptavidin四聚体的最高耐受温度仅为65ºC，因此，Traptavidin的热稳定性更好(图1B)。
