细胞培养的环境参数对细胞的生长状态至关重要,需严格控制以下条件:温度:维持在37℃,模拟哺乳动物体内环境。温度过高或过低均会影响细胞的代谢活动和增殖速率;CO2浓度:保持在5%。CO2与培养基中的碳酸氢钠缓冲系统共同作用,维持培养基pH值在7.2-7.4之间,确保细胞处于适宜的酸碱环境中;湿度:培养箱内湿度需保持在95%以上,以防止培养基蒸发导致渗透压变化。
