RNase A is an endonuclease that cleaves the phosphodiester bond of single stranded RNA after pyrimidine nucleotides. It can cleave the 3 'phosphate end (e.g. pG-pG pC-pA pG will cleave into pG-pG pCp and A-pG). It exhibits the highest activity towards single stranded RNA. RNase A is a single chain polypeptide containing four disulfide bonds. It is different from RNase B and is not a glycoprotein. Ribonucleases do not hydrolyze DNA because DNA lacks the necessary 2 '- OH groups to form cyclic intermediates. RNase A can also hydrolyze RNA in protein samples. RNase A can be inhibited by the alkylation of His12 and His119 and activated by potassium and sodium salts. RNAse is inhibited in the presence of heavy metal ions. In addition, RNase is also competitively inhibited by DNA。
