Poly ADP-ribose (PAR) is a polymer formed by the repetitive addition of ADP-ribose units to proteins, a process known as ADP-ribosylation. This modification is catalyzed by poly(ADP-ribose) polymerases (PARPs), particularly PARP-1, in response to DNA damage. PAR plays a crucial role in regulating DNA repair, genomic stability, and cell survival by modifying target proteins involved in DNA repair processes and cellular stress responses. The formation of PAR is also involved in transcription regulation, chromatin remodeling, and apoptosis. Due to its involvement in cellular response to stress, inflammation, and cancer, poly ADP-ribose is a key area of research in DNA repair mechanisms and therapeutic interventions targeting PARPs in diseases like cancer.
