Presenilin-1 (PSEN1) is a key protein involved in the processing of amyloid precursor protein (APP), a process that is central to the development of Alzheimer's disease. It is a catalytic subunit of the γ-secretase complex, which cleaves APP to produce amyloid-β peptides. Accumulation of these amyloid-β peptides in the brain is a hallmark of Alzheimer's disease. Mutations in the PSEN1 gene are strongly associated with early-onset familial Alzheimer's disease, leading to an abnormal increase in amyloid-β production. Besides its role in APP processing, presenilin-1 is also involved in notch signaling, which is important for cell fate determination, neural development, and cellular differentiation.
