Tyrosine Hydroxylase (TH) is an enzyme that catalyzes the rate-limiting step in the synthesis of catecholamines, including dopamine, norepinephrine, and epinephrine. It converts the amino acid tyrosine into L-DOPA, which is subsequently decarboxylated to form dopamine. This enzyme plays a central role in the nervous system, particularly in dopaminergic neurons, where it is essential for the production of dopamine, a neurotransmitter involved in regulating mood, motor control, and reward systems. Tyrosine hydroxylase is primarily expressed in the brain, adrenal glands, and sympathetic nervous system. Dysregulation of TH activity is associated with several neurological disorders, such as Parkinson's disease (where dopamine-producing neurons are lost) and drug addiction (due to its involvement in the reward pathway).
