法合成环肽，尤其是用于中环肽的合成。固相合成常用MBHA，Oxime 和PAM 等树脂。氨基酸的侧链（如Asp，Glu 的侧链羧基；Ser，Thr等的侧链羟基）及羧端均可通过缩合反应固载在树脂上，若按氨基酸的C端固定在树脂上的方式可以分为三种：(1)头/尾锚定。一般利用C端的羟基将肽链结合在树脂上，然后脱去N端保护，游离的氨基进攻接在树脂上的C末端，成环的同时目标环肽从树脂上脱下。（2）侧链锚定。主要是将肽链的侧链连接在树脂上，待目标环形成后用相应化学方法脱下。 Figure
