α-氨基酸之一。 L化合物为明胶、麸蛋白（麦醇溶蛋白）、玉米醇溶蛋白、鲱精蛋白、鲑精蛋白、酪蛋白等多种蛋白的组成成分，特别是明胶中最多（约占 20％）。是唯一的醇性氨基酸，易溶于水，茚满三酮反应为黄色，由古柯叶液碱（ hygrine，属氮杂茂衍生物的生物碱）氧化所得的古柯叶液碱酸为脯氨酸的甲基衍生物，植物中存在的脯氨酸二甲内盐是二甲 -L-脯氨酸（甜菜碱）。为非必需氨基酸，在生物体内经谷氨酸被代谢。
可将二肽水解成二个氨基酸的酶。在不同的生物中具有其不同的特异性。现在知道有对应于甘氨酰-甘氨酸、甘氨酰-L-亮氨酸、X－L组氨酸、L-半胱氨酰-甘氨酸、L-脯氨酰-X（脯酰肽酶）、X－L-脯氨酸（脯氨肽酶）（X=氨基酸残基）等的二肽酶。
