CAR-T细胞的发育是一个复杂的过程,需要i)筛选和测序肿瘤抗原特异性单克隆抗体(mab);II)合成单链抗体(scFv)cDNA,并将其克隆到慢病毒载体的嵌合抗原受体(CAR)盒中(如慢病毒载体的第3代CAR盒中的抗bcma单链抗体);III)高滴度慢病毒CAR编码慢病毒的包装和生产;IV)分离、激活和扩增具有特定细胞表型(如CD4+或CD8+或混合细胞)的患者来源的t细胞;v)并用CAR编码慢病毒转导活化的t细胞;VI)流式细胞术和功能分析验证工程化CAR-t细胞。BPS建立了抗bcmaCARJurkat/nfat荧光素酶稳定报告细胞系,是利用CAR-t慢病毒和Jurkat/nfat荧光素酶报告细胞系进行的一系列报告生物测定之一。抗bcmaCARJurkat/nfat荧光素酶报告细胞系是预测抗bcmaCAR慢病毒作用机制和治疗潜力的重要系统。它是用抗bcma单链抗体CAR慢病毒(BPSBioscience#79701)转导Jurkat/nfat荧光素酶报告细胞而形成的单细胞克隆稳定细胞系。
