Percoll is composed of silica gel particles with a particle size of 15-30nm (23% w/w in water), which are coated with polyvinylpyrrolidone (PVP). It is used to establish non-toxic, low viscosity, and low osmotic pressure density gradients, which can be used for separating cells, organelles, and viruses. It has been used to separate the inner and outer membrane vesicles of Escherichia coli. Percoll is isotonic, non-toxic, and easily removed from purified materials throughout the entire gradient process. Characteristics and advantages Low osmotic pressure allows for precise adjustment of physiological conditions without significant interference from the medium. Compatible with live cells and viruses, and allows for the isolation and recovery of complete, fully active systems. It is impermeable to biofilms and does not change the buoyancy density of particles during centrifugation. The gradient spontaneously formed during centrifugation allows for the mixing of a large amount of samples in the centrifuge tube. Low viscosity leads to rapid gradient formation and particle separation. Most biological particles with a settling coefficient greater than 60S can be successfully separated in Percoll gradient
