    血小板生成素（Tpo）是巨核细胞生成和血栓形成的关键调节因子。
    它主要在肝脏中产生，并被受体 Tpo R/c-mpl 结合和内化。Tpo-Tpo-R
    信号通路的缺陷与多种血小板疾病有关。353 个氨基酸（aa）的人 Tpo
    前体被切割以产生 332 个氨基酸的成熟蛋白。成熟的人 Tpo 与小鼠和大
    鼠 Tpo 具有大约 70%的 aa 序列同源性。它是一种 80-85kDa 的蛋白质，
    由一个与促红细胞生成素（Epo）同源的 N 末端结构域和一个包含多个
    N 连接和 O 连接糖基化位点的 C 末端结构域组成。Tpo 促进 MK 及其
    祖细胞的分化、增殖和成熟，其他几种细胞因子也可以促进这些功能，
    但只能与 Tpo 协同作用。Tpo 还能促进血小板的产生、聚集、ECM 的
    粘附和活化。它在 C 末端结构域内被 Arg191 之后的血小板衍生的凝血
    酶切割，随后在扩展消化时在其他位点被切割。全长 Tpo 和较短的形式
    在等离子体中循环。C 末端结构域不是结合 Tpo R 或诱导 MK 生长和分
    化所必需的。除了造血作用外，Tpo 在大脑中表达，促进缺氧致敏神经
    元的凋亡，并通过阻断 NGF 诱导的信号传导抑制神经元分化。血小板
    生成素受体（Tpo R），也称为骨髓增殖性白血病蛋白（c-Mpl），是一
    种 I 型跨膜蛋白，是造血素/细胞因子受体超家族的成员。作为选择性剪
    接的结果，有四种已鉴定的信使核糖核酸变体。该功能性受体由信使核
    糖核酸的 P 亚型编码。人类和小鼠受体共享大约 81%的氨基酸序列同一
    性。Tpo R 在各种细胞类型中低水平表达，包括造血祖细胞、巨核细胞
    和血小板。
