文献和实验. Test each tube w/H2O and discard.Add protein mixture to dialysis tube. Stir slowly in 100 mM Tris buffer or Binding buffer until pink color is out.Transfer protein mixture (Ab) to 50 ml Falcon tubes to determine volume.Dilute the mixture
(collect at ~5 min/fraction). 25 tubes are sufficient for collection. In general, you can see high Ab concentrations in 7-8 tubes. Test 1 ul on pH paper. Read OD to know Ab concentrations. OD (Absorbance at 280 nm)/1.35 = X
1 well for each of the 1º Ab as single positive controls for setting compensation. Incubate 60 min at 4ºC. Spin 1500 RPM x2 min. Discard supernatant by shaking it out once into the sink, and blot inverted plate on paper towel. Wash by adding 200 uL
