稳定性
热稳定性以损失率显示。损失率是由加速降解试验决定,具体方法如下:在37°C孵育48小时,没有显著的降解或者沉淀产生。保质期内,在适当的条件下存储,损失率低于5%。

腹水。刚采出的腹水含有大量的油脂、巨噬细胞、杂交瘤细胞、腹腔内的上皮细胞、血液中的细胞成份等杂质,尤其是这些细胞成份,如果不除去则可能在腹水的长期保存中慢慢破裂,细胞内的蛋白质成份会释放到腹水中,增加了腹水中的杂质,使纯化变得困难,因此腹水在采集后不管是要长期保存还是立即纯化都应该先离心除去细胞的成份,同时也去掉油脂(油脂的密度比较小,一般都漂浮在腹水表面,用枪吸出丢掉即可)。单抗的纯化目前主要有这么几种: (1)盐析/沉淀法; (2)离子交换法;(3)凝胶过滤层析;(4)蛋白A或蛋白G纯化
的检测抗原是否纯净而定。如杂交瘤抗体是针对纯化的蛋白质抗原的,100%的抗原参与反应,一个阳性/阴性判别系统就够了。另一方面,如果杂交瘤抗体是针对细胞表面微量的蛋白抗原,检测系统可能需要能测出微弱信号,则动力学范围至少应为10:1,最好为100:1。另外,检测方法的选择还受所需杂交瘤抗体的类型和预定的用途的影响。结合补体的抗体可以用基于细胞毒性反应的检测方法来选出。如需结合A蛋白的杂交瘤抗体,就要用结合蛋白A的检测方法。 下面简要介绍几种常用的抗体检测方法: ⑴ 免疫
一、免疫亲和层析程序方案概述 1. 将抗体与基质(微珠)共价交联并制备抗体亲和层析柱。 先将单克隆抗体或经过亲和层析纯化的多克隆抗体以共价方式交联到固相基质(微珠)上。除特殊情况外,一般不使用未经纯化的多抗,因为从多抗层析柱上洗脱抗原非常困难,通常需要以适当的条件先使抗原变性。抗体以共价方式交联到固相基质上有很多不同的方案,将抗体交联到蛋白A或蛋白G微珠上可能最简单。另一种方法是将抗体共价交联到具有活性基团的微珠上。 2. 将抗原结合到抗体-微珠基质上。 在抗体-微珠基质
