牛转铁蛋白(饱和铁形态,细胞培养级别)转铁蛋白冻干粉需要保存在冷藏状态下,使用之前再溶解。一般用PBS或者其它合适的溶剂在室温下将转铁蛋白冻干粉溶解成储液,浓度在0.5-2.0mg/ml,然后用无菌滤膜进行过滤,滤液可在无菌条件下2-8℃保存30天。对于杂交瘤,黏连细胞和悬浮细胞系,在细胞培养过程中,应保持培养基中转铁蛋白浓度在0.5-100μg/ml。针对不同的细胞系,需要选择最合适的浓度。
牛转铁蛋白(饱和铁形态,细胞培养级别)转铁蛋白是血浆糖蛋白,从血浆中分离而来,牛转铁蛋白以两种分子形态存在,分子量分别为74KD和78KD。每个转铁蛋白分子通过C端和N端结构位点可以结合两分子铁离子,所以转铁蛋白以三种形态存在:脱铁(不结合铁离子),饱和铁(结合两分子铁离子)和部分饱和,在生理情况下,转铁蛋白分子仅有 1/3 被铁饱和。转铁蛋白对于绝大部分细胞系在无血清培养基中生长是必需的,研究表明饱和铁形态的转铁蛋白对于促进细胞生长是最有效的形态。
