产品介绍：Alpha 1 Fetoprotein is a major plasma protein produced by the yolk sac and the liver during fetal life. Alpha fetoprotein expression in adults is often associated with hepatoma or teratoma. However, hereditary persistance of alpha-fetoprotein may also be found in individuals with no obvious pathology. The protein is thought to be the fetal counterpart of serum albumin, and the alpha fetoprotein and albumin genes are present in tandem in the same transcriptional orientation on chromosome 4. Alpha fetoprotein is found in monomeric as well as dimeric and trimeric forms, and binds copper, nickel, fatty acids and bilirubin. The level of alpha fetoprotein in amniotic fluid is used to measure renal loss of protein to screen for spina bifida and anencephaly. Expression has been documented in human adrenal, liver, ovary, testis, and pancreas. ESTs have been isolated from normal human brain, liver/spleen, embryo and uterus tissue libraries.

二抗的4个基本步骤：
1. 选择正确的种属来源和反应物种
二抗的选择首要基于一抗，应与一抗的种属来源相对应，且需要来自于不同的物种（如下图所示）。

2. 选择正确的二抗形式
选择合适且正确的二抗形式，也是获得实验成功的关键之一。三种二抗形式Whole IgG、F(ab') 、Fab如下图所示。

3. 抗体特异性的选择
要注意区分二抗的特异性和形式的不同，特异性指的是二抗识别的不同一抗亚型或某特殊区域。抗体的特异性主要基于以下三种形式：
（1）轻链特异性
（2）Fc端特异性
（3）亚型特异性

4.偶联标记的选择
不同实验类型适合的标记类型不一样，如下表所示：

免疫荧光双标抗体选择注意事项：
双重免疫荧光
    在同一组织或细胞标本同时检测两种抗原时，需进行双重荧光染色。双重免疫荧光标记法也分为直接法和间接法。
直接法双重免疫荧光标记
   直接法双重免疫荧光标记：将标记有两种不同荧光素的抗体（如抗A和抗B）以适当比例混合，滴加在标本上孵育，然后洗去未结合的荧光抗体，在荧光显微镜下分别选择两种相应的激发滤片观察，即可对两种抗原进行定位和定量。直接法简便可靠，但灵敏度较低。
间接法双重免疫荧光标记
   间接法双重免疫荧光标记：用未标记的两种特异性一抗（不同种属来源）孵育组织或细胞，洗去多余的一抗后，再用两种不同的荧光素分别标记的二抗孵育组织或细胞，洗去多余的二抗体，后在荧光显微镜下分别选择两种相应的激发滤片观察，从而对两种抗原进行定位和定量。
