MicroRNAs是一类非编码蛋白的RNA片段,长度大约为22个核苷酸。它是通过结合到目标mRNA上进而抑制该mRNA翻译来起作用的。由此看来，MicroRNAs无论在健康的还是疾病的机体中，都起着中枢调控基因功能的作用。当前，222种人类的MicroRNAs已经被发现：86种通过随机克隆和测序的方法，43种通过计算机的方法，剩下的则是推测的其他生物体中MicroRNAs的类似物。为了验证我们的设想，即microRNAs的总体数量可能更多，而且一些特异性地存在于灵长类动物中
个，20例标本中共存性差异表达miRNA有9个（上调2个，下调7个，P〈0．01）；选取表达明显上调的miR-222作为进一步研究对象，半定量RT—PCR和Western blot分别证实其调节的靶目标-结缔组织生长因子（CTGF）在mRNA水平HCC和癌旁组织中差异无统计学意义。而蛋白质水平在HCC中表达明显下降。结论液相芯片筛选差异表达miRNA为HCC发病机制和诊断治疗的研究提供了新的思路；miR-18可能通过转录后基因沉默机制抑制CTGF mRNA翻译，从而促进HCC浸润转移。 【关键词】癌
